The two Florida-based teams met at the DRV PNK Stadium in the round of 32 of the Leagues Cup.
And Messi's side booked their spot in the next round of the competition following their 3-1 victory.
The seven-time Ballon d'Or winner bagged a brace, with Josef Martinez also getting on the score-sheet.
He now has five goals in three matches for his new side as he enjoys a prolific start to his career in America.It wasn't just the goals that impressed fans online as the 36-year-old also showed his intensity and the passion he has for the game.
Messi was involved in several flashpoints during Inter Miami's fixture against Orlando City as he clashed with the opposition, including a bust-up in the tunnel.
He was also involved in an angry exchange with the game level at 1-1 and reacted furiously to a foul, waving an imaginary yellow card to the match referee.
What's more, the former Barcelona man unleashed another side to his character to the amazement of fans on social media.
With Inter Miami leading 3-1, Messi clashed with Brazilian Felipe Martins as they angrily exchanged words before Leonardo Campana intervened.Messi's intensity and being dialed in for the derby match sent social media wild.
One Twitter user wrote: "Alba and Busquets wondering what the hell happened to him since he left Barcelona."
